- W2008617103 abstract "RATIONALE: Hymenoptera venom is composed of allergen protein components including phospholipase and hyaluronidase. Vespidae species also contain the protein Antigen 5. For US venom allergen extract products, FDA standardization includes measuring the phospholipase and hyaluronidase enzyme activity and labeling by total protein content. Antigen 5 is an important allergen with a high prevalence of IgE binding, but the content of this protein is not a requirement for FDA standardization. Since Antigen 5 has no defined enzyme function, electrophoresis can be used to identify it in venom products. The purpose of this study was to use rVes v 5 to localize this allergen in SDS-PAGE gels and estimate the concentration in US products. METHODS: US commercial allergenic venom extracts of Vespula/Dolichovespula (Yellow jacket, White faced hornet, Yellow hornet), Polistes (Wasp), and Honeybee were studied using SDS-Gel electrophoresis. Lyophilized purified recombinant Ves v 5 protein was reconstituted to 180μg/mL, serially diluted twofold to 5.6μg/mL, and run on the same gels. The gels were stained for protein and analyzed using gel analysis software. RESULTS: All venom batches except for Honeybee showed the presence of Antigen 5 type allergen protein on SDS-PAGE gels. The intensity of the gel band in the venoms matched the 10-30μg/mL dilution of the rVes v 5. Since the extract samples were formulated to 100μg/mL, the Antigen 5 proteins are 10-30% of the venom proteins. CONCLUSIONS: This study demonstrated that US venom products used for diagnosis and immunotherapy contain the important allergen Antigen 5." @default.
