- W2008848604 abstract "A breakdown of the tryptophan metabolism seems to play an important role in the etiology and the pathogenesis of Major depression. Several studies demonstrated changes in the kynurenine pathway, the metabolism of tryptophan to kynurenines, in blood of patients with affective disorders. These studies led to the hypothesis of increased microglial synthesis of the kynurenine metabolite quinolinic acid (an endogenous NMDA-receptor agonist) in depressed patients. Myint et al. (2007) presumed a breakdown of the neuroprotective – neurodegenerative balance because of this increase. However, no study of human brain tissue has been published on this topic yet. To clarify this question we evaluated the quantity and cell density of quinolinic acid-immunopositive (QA+) microglia in the pregenual as well as in the superior and inferior postgenual anterior cingulate gyrus (CG). We analyzed brain tissue from 7 patients with major depressive disorder, 7 patients with bipolar disorder and 10 matched controls from the Magdeburg brain collection. ANOVA showed no effect of diagnosis on quantity or cell densitiy of QA+ microglia. However, the cell density of QA+ microglia was increased in the pregenual anterior CG of patients with bipolar disorder in comparison to controls. This result will be reassessed by analyzing a higher number of cases. In addition, single case analysis may reveal further information on the dependence of results from disease severity or psychopathologic subtypes." @default.
