- W2009371708 abstract "To determine if manganese (Mn) undergoes presystemic elimination, male Sprague-Dawley rats were administered 54MnCl2 in doses of 0.1, 0.3, 1.0, and 3.0 mg of Mn/kg into either their ileocolic (portal) or femoral (systemic) vein. Blood and bile samples were obtained for up to 90 min after administration. The area under the plasma concentration versus time curve (AUC) was determined for each route of administration. A ratio of 0.65 to 0.7 for the portal AUC relative to the systemic AUC indicates that systemic availability of Mn following portal administration is approximately 30 to 35% less than that after systemic administration of low doses of Mn. Concentration of Mn in liver was greater after portal administration of 0.1 mg Mn/kg than after systemic. Biliary excretion of Mn was also enhanced by portal administration relative to systemic, although the transfer from liver to bile seemed less affected by the route of administration than the transfer from blood to liver. Infusion of Mn by either route demonstrated that more Mn was required to produce death when given portally than when given systemically. In summary, Mn undergoes significant presystemic elimination. This effect appears to be dose dependent and may serve a protective function against acute Mn toxicity." @default.
