The method employs a water-soluble singlet oxygen source and a superoxide-detecting reagent such as a combination of p-nitrotetrazolium blue and superoxide dismutase. The generation of the superoxide ion from oxidation of hydrogenated nicotinamide adenine dinucleotide and 5-hydroxytryptophan with singlet oxygen was confirmed by this method.
