- W2010414303 abstract "ABSTRACT Microbial assay for detection of residual levels of furazolidone (FZD) residues in cultures after cold storage and thermal processing was developed. Bacillus subtilis (G-2), a sensitive microflora isolated from the shrimp gut, was used as the test organism. The assay had a linear relationship with a positive correlation of 0.884 over a range of 1 to 100 μg/ml concentrations. Shrimp spiked with 5 to 100 μg/ml of FZD and extracted using acetonitrile showed 77.9% recovery. Stability of FZD residues to cooking (100°C for 10, 20, and 30 min), retorting (121°C for 10 and 15 min), iced (0°C), and refrigerated (5°C) storage was examined. The loss of FZD to cooking was 9%, 15%, and 20%, respectively. The loss was 26% and 27% after retorting for 10 and 15 min, respectively. The loss was high in iced stored shrimp with 28%, and in refrigerated shrimp with 35%. The degradation was higher in cold stored shrimp than thermally processed shrimp. The developed assay shall therefore be very useful for screening the presence of FZD residues in cold stored and thermally processed shrimp, as there is zero tolerance for FZD in the market.
