- W2010459548 abstract "The application of flow cytometry on telomeric length determination was initially described by Hultdin et al. (1) and Rufer et al. (2) in 1998. Since then similar techniques have been used for the study of telomere length regulation in immunobiology(3-5), transplant biology (6, 7), and hematological disorders (8, 9). Recently, we published a paper reporting the validation and development of quantitative flow cytometric technique in our institute (10). In their letter, Roos and Hultdin emphasised on the need of appropriate internal control and the importance to consider the DNA index (DI) of each sample for correct interpretation of telomere length. We agree fully with the idea of Roos and Hultdin. Their additional information on MOLT-4 actually matched with our description. We have demonstrated that by taking into account the tetraploid nature of MOLT-4 cells, we were able to explain why the Flow FISH fluorescence value obtained was doubled that predicted by our “conversion” curve. Even though K562 has been described as a diploid cell line from our source (American Type Cell Culture), a “correction factor” of 2.32 would be needed for the Flow FISH fluorescence value to be moved back on the “conversion” curve. Even if our Flow FISH fluorescence value for K562 is compensated by the DI provided by Roos and Hultin (DI = 1.46), it is impossible to explain the high fluorescence value obtained by Flow FISH. The correction of Flow FISH fluorescence by DI has not been described in other groups mainly because normal cells were being studied. We agree that DNA analysis must be included in study of true aneuploid and polyploid cells. With more recent development, it is possible to analyse the cell surface markers and telomere length simultaneously by flow cytometry (4, 11).
