- W2011019715 abstract "188 Numerous animal models have been used in efforts to describe in vivo events responsible for allograft rejection. However, interspecies differences in immune responses have made clinical correlates with the human transplant experience difficult to establish. Recently, the severe combined immunodeficient mouse (SCID), skin grafted and reconstituted with human peripheral lymphoid cells has been shown to provide an in vivo environment capable of mediating alloimmune destruction of human dermal micro vessels. We have extended those observations to evaluate human immune injury in a vascularized arterial allo- transplant model. Under nembutal anesthesia and 40× magnification, 4-6mm segments of size matched, fresh human artery were used to replace similar length segments of CB.17SCID/beige mouse aorta. Human peripheral blood lymphocytes (PBL) were obtained from normal human volunteers. Seven days post transplant, 2-3 × 108 human PBL were administered to experimental animals i.p. Fourteen days later, the percentage of circulating human CD3+ cells were determined by flow cytometry, and lymphoid reconstitution was considered successful when a discreet population of human CD3+ lymphocytes was identified. Forty five of 48 (93%) transplanted mice survived, including 14 control animals with arterial transplants who did not receive PBL, 4 animals who received PBL but failed to demonstrate circulating human CD3+ cells, and 27 who received human PBL and demonstrated circulating human CD3+ cells. Surviving animals were sacrificed 14, 21 or 28 days after PBL reconstitution, and recovered arteries were analyzed by histology and immunohistology. Each of the 14 control animals demonstrated a patent human arterial graft with normal vascular histology, and without lymphoid infiltration. The results from transplanted and reconstituted mice have been summarized below: Table Additionally, 10 of 10, 28 day mice displayed CD3+ cellular infiltration when frozen section immunohistology was performed.
