Inspection of hamster embryos 10 hr after a teratogenic dose of sodium arsenate revealed a delay in neural-fold elevation and neural-tube closure. There was a paucity of cephalic mesoderm in treated embryos as compared to control embryos of an equivalent gestational age. The results suggest that arsenic-induced damage to the paraxial mesoderm may be related to the ultimate skeletal malformations. Intravenous injection of 20–100 mg/kg of methylarsonic acid or 20–100 mg/kg of dimethylarsimic acid, the methylated metabolites of inorganic arsenic, failed to induce the marked teratogenic response that was associated with injection of the inorganic arsenic compounds. The results suggest that in situ conversion of inorganic arsenic to methylated metabolites represents a detoxication pathway for both the dam and the developing embryo." @default.
