- W2012393160 abstract "Summary Cells of Scenedesmus obtusiusculus Chod. were grown in a phosphorous-free nutrient medium for 48 hours and then used for experiments concerning the effects of readdition of phosphate on energy metabolism. Addition of 0.25 mM phosphate, which is saturating for uptake, approximately doubled the respiratory oxygen consumption. This effect was accompanied by a steady rise in the cellular ATP pool. Both the stimulation of oxygen consumption and the increment in ATP was sensitive to antimycin A. Under dark anaerobic conditions, phosphate addition caused a transient peak in ATP. Presence of phosphate strongly affected the pattern of ATP pool fluctuations in dark-light and light-dark transitions. Net O 2 evolution in the light was decreased by phosphate addition. Providing phosphate at steady state conditions in the light in a N 2 atmosphere produced a rapid drop in the ATP content, followed by a recovery. In cells treated with DCMU (1 and 50 μM), addition of phosphate was accompanied by a rapid and permanent increase in the ATP pool. In the presence of 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIB) or phloridzin at concentrations that block photophosphorylation, transient peaks in the ATP level occurred with maxima after 1 min.
