- W2012462398 abstract "RATIONALE: We evaluated whether bodies and feces from D. pteronyssinus and D. farinae house dust mite (HDM) species are both needed for manufacturing desensitization extracts. METHODS: We performed an allergomic characterization of bodies and feces purified from D. pteronyssinus and D. farinae. Potential contamination of whole cultures of both species by residual medium components was evaluated using Western blot and mass spectrometry. RESULTS: In D. pteronyssinus, bodies contain high amounts of group 2 major allergen as well as group 20 allergen, while feces are the main source of group 1 relevant allergen as well as group 5, 8-10, 13 and 21 allergens. In D. farinae, bodies are enriched in group 3, 14 and 20 allergens, while feces contain the group 1 relevant allergen as well as group 5, 7, 9, 13, 21 and 22 allergens. We confirm that whole cultures from both species obtained on a defined medium free of animal components are suitable for manufacturing pharmaceutical-grade extracts. Indeed, such cultures are free of contamination by proteins from the medium. A seroepidemiological study conducted in more than 600 North American and European HDM-allergic patients confirms the need for immunotherapies with well balanced allergen contents, most particularly as it relates to Der p 1, Der p 2, Der f 1 and Der f 2. CONCLUSIONS: For both D. pteronyssinus and D. farinae, bodies and feces display distinct allergen repertoires which complement each other in view of the sensitization profiles of HDM-allergic patients.
