- W2012606784 abstract "Abstract The role of monocytes and prostaglandin E (PGE) in control of MS lymphocyte adherence to measles virus-infected human epithelial cells was examined. Adherence of control and MS lymphocytes was significantly reduced when monocytes were removed from mononuclear cell suspensions by adherence to glass, to tissue culture cell microexudate-coated flasks, and by ingestion of iron filings. Adherence to uninfected cells was not altered by monocyte depletion. Lymphocyte adherence determination (LAD) values could be restored by incubation of depleted cell suspensions with PGE 1 and E 2 or by adding back monocytes at a 20:1 or 10:1 lymphocyte to monocyte ratio. These results indicated that monocytes, perhaps by virtue of their capacity for PG synthesis, are required for optimal adherence of lymphocytes to virus-infected cells. MS monocytes produce significantly higher levels of PGE in tissue culture than do control monocytes, and monocyte-depleted MS leukocytes are more sensitive to PGE-induced increases in LAD at lower concentrations of PGE (10 −12 M ) than control lymphocytes. MS mononuclear cells pretreated with 5 × 10 −5 M aspirin, were more sensitive to PGE-induced increases in LAD at concentrations from 10 −4 –10 −14 M . Thus increased lymphocyte adherence in MS patients appears to result from defective monocyte-mediated control and abnormal lymphocyte responses. The cell-cell interactions responsible for the in vitro test may be regulated by prostaglandins." @default.
