The haemoglobin binding capacity of the serum was estimated in 64 normal adults, 83 healthy elderly persons, 73 patients suffering from various forms of congenital and acquired haemolytic anaemia, 29 patients with non-haemolytic anaemia, 28 patients with pnncytopenia, 4 patients with pernicious anaemia and 67 patients suffering from various disorders not associated with anaemia, and in whom increased haemolysis could not be demonstrated. In a group of 41 patients and one normal control a comparative study was made of the results obtained by this technique and by the determination of the erythrocyte survival using radioactive chromium. Haptoglobin levels are of value in detecting the occurrence of increased haemolysis, provided the erythrocyte sedimentation rate is taken into account in evaluating thc results, as a raised E.S.R. is usually associated with a raised haptoglobin level. The teclinique used for the determiuation of haptoglobin (electrophoresis on cellulose acetate strips) is described in detail. In view of its simplicity, it would appear to be of practical value in the differential diagnosis of anaemias and jaundice. Résumé Le rapport entre l'hémolyse augmentée et le taux d'haptoglobine sérique a été étudié. La faculté de l'haptoglobine de lier l'hémoglobine du sérum a été mesurée chez 64 adultes normaux, 83 personnes âgées en bonne santé, 73 patients souffrant de diverses formes d'anémies hémolyticpes congénitales ou acquises, 29 patients souffrant d'anémies non hémolytiques, 28 patients atteints de pancytopénie, 4 patients atteints d'anémie pernicieuse et 67 patients souffrant de troublcs divers non associés A une anémie et chez lesquels une hémolyse augmentée n'a pas pu être démontrée. Daus un groupe de 41 patients et un groupe de contrôle ronstitué de personnes normalcs, il a été procédé B une étude comparative entre les résultats obtenus par cette teclinique et les résultats obtenus par la détermination de la survie des érythrocytes marqués au radiochrome. La mesure du taux de l'haptoglobine est valable pour déterminer la présence d'une hémolyse augmentée, mais il faut tenir compte de la vitesse de sédimentation dans l'estimation des résultats puisque une vitesse de sédimentation auginentée est en général associée à une augmentation du taux de I'haptoglobine. La technique utilisée pour le dosage de l'haptoglobine (électrophorèse sur bande de cellulose acétate) est décrite en détail. Etant donné sa simplicité, il semble que cette technique a unc valeur pratique pour étayer le diagnostic différentiel des anémies et des ictères. Zusammenfassung Es wurde eine Untersuchung über die Beziehungen zwischen einer gesteigerten Hämolyse und dem Serum-Haptoglobinspiegel durchgeführt. Die Hämoglobin-bindungskapazität des Serums wurde bei folgenden Personen bestimmt: 64 gesunde Erwachsene, 83 gesunde ältere Personen, 73 Patienten mit verschiedenen Formen von angeborener oder erworbener hämolytischer Anämie, 29 Patienten rnit nicht hämolytischer Anämie, 28 Patienten mit Pancytopenie, 4 Patienten mit perniziöser Anämie und 67 Patienten mit verschiedenen Leiden, aber ohne Anämie und ohne nachweisbare gesteigerte Hämolyse. In einer Gruppe von 41 Patienten und einer gesunden Kontrollperson wurde eine vergleichende Untersuchung mit der vorliegenden Technik und der Erythrozytenlebensdauerbestimmung mit radioaktivein Chrom durchgeführt. Die Haptoglobinspiegelbestimmung ist ein wertvolles Hilfsmittel zur Erkennung von Zuständen mit gesteigerter Hämolyse. Dies gilt allerdings nur unter der Voraussetzung, daß die Blutsenkungsgeschwindigkeit mit berücksichtigt wird.
