- W2013753453 abstract "This chapter discusses the radioassay for dihydropteroate-synthesizing enzyme activity. Dihydropteroate-synthesizing enzyme catalyzes the coupling reaction of hydroxymethyldihydropterin or its pyrophosphate ester and p-AB to form dihydropteroic acid in the presence of both adenosine triphosphate (ATP) and Mg2+ or Mg2+ alone as cofactor. Because dihydropteroic acid has folic acid activity for microorganisms, the activity of this enzyme has been measured, by microbiological assay, with Streptococcus faecalis R. The enzyme is widely distributed in higher plants and high enzyme activity is also found in green leaves. The simple and rapid radioassay procedure discussed is based on the direct measurement of the formation of 14C-labeled dihydropteroic acid from 14C-labeled p-AB and pyrophosphorylmethyldihydropterin. The labeled dihydropteroic acid, produced by the enzymic reaction, is separated easily and satisfactorily from the labeled substrate by paper chromatography and the radioactivity of the paper section, containing the labeled product, is counted in a liquid scintillation counting system. By applying the radioassay procedure, the distribution of dihydropteroate-synthesizing enzyme in several higher plants has been investigated and the results are described in the chapter." @default.
