- W2014113467 abstract "Mycoplasma (M.) mobile 163 K, isolated from the gills of a tench (Tinea tinea L.), was examined for cytotoxic capacities using tracheal organ cultures from gnotobiotic rats and gnotobiotic piglets in pH-controlled experiments (pH 7.2–7.6). The mycoplasmas caused an inhibition of the ciliary activity at incubation temperatures of 20, 25 and 30°C. The strongest cilia stopping effect was observed at an incubation temperature of 25°C, the optimal growth temperature of the mycoplasmas. No ciliostasis occurred at 37°C. The number of the organisms did clearly affect the severity of ciliostasis in the range from 4.0 × 104 to 7.7 × 108 c.f.u. in rat as well as in porcine tracheal organ cultures. Toxic substances, secreted by the mycoplasmas into the culture medium, could not be detected. In histological investigations cytopathological changes were observed in the epithelial cells, apparent in the destruction and loss of cilia, cytoplasmatic vacuolization, swelling of mitochondria, peripheral orientation of the nuclear chromatin and detachment of epithelial cells from each other and the basal membrane. The final stage of the infection was characterized by complete exfoliation of the epithelial cells and the complete destruction of the multi-layer epithelium. The localization of the mycoplasmas attached to the ciliary epithelium was shown by scanning and transmission electron microscopy. In der vorliegenden Untersuchung wurde der Mykoplasmenstamm Mycoplasma (M.) mobile 163 K, der von den Kiemen einer Schleie (Tinea tinca L.) isoliert wurde, auf cytotoxische Eigenschaften in Trachealorgankulturen von gnotobiotischen Ratten und gnotobiotischen Schweinen geprüft. Die Untersuchungen erfolgten in pH-Wert kontrollierten Medien (pH 7,2–7,6). Die Mykoplasmen hemmten die Cilienaktivität bei 20, 25 und 30°C. Der stärkste Effekt war bei 25°C zu beobachten, der optimalen Wachstumstemperatur von M. mobile. Bei 37°C kam es nicht zur Hemmung der Cilienaktivität. Die Zahl der Organismen hatte sowohl in den Ratten- als auch in den Schweinetrachealkulturen im Bereich von 4,0 × 104 bis 7,7 × 108 CFU einen deutlichen Einfluß auf die Ciliostasis. Toxische Substanzen waren im Kulturmedium der Mykoplasmen nicht nachzuweisen. In den histologischen Untersuchungen konnten folgende cytopathologische Veränderungen in den Epithelzellen beobachtet werden: Zerstörung und Verlust der Cilien, cytoplasmatische Vakuolisierung, An-Schwellung der Mitochondrien, periphere Verlagerung des nuklearen Chromatins. Die Epithelzellen lösten sich voneinander und von der Basalmembran. Das Endstadium der Infektion war durch eine vollständige Zerstörung des mehrschichtigen Epithels charakterisiert. Die Lokalisation der an das Cilienepithel adhärierten Mykoplasmen wurde im Raster- und Transmissionselektronenmikroskop dargestellt." @default.
