[2] that inulin appears to be eliminated by the kidney at the same rate as fluid is filtered by renal glomeruli, measurement of inulin clearance was established as the reference clinical method for estimating glomerular filtration rate. Gaudino et al. [3] later demonstrated that inulin can also be used to measure extracellular fluid space. Although [t4C]and [3H]inulin are available to facilitate measurements of glomerular filtration rate [4] and extracellular fluid space [5] in experimental animals, clinical studies with inulin have been impeded by the lack of satisfactory methods to measure concentrations of nonradioactive inulin in biological fluids. Classical calorimetric methods entail initial hydrolysis of inulin with subsequent quantitation of the fructose that is formed. Since other compounds react with the chromogenic reagents that have been used [6, 71, analytical sensitivity is limited and precision at low levels is suspect because the assumption is made that the initial blank readings represent the level of interference in all subsequent samples. Accordingly, we developed a high-performance liquid chromatographic method for measuring inulin concentrations in plasma and urine." @default.
