The study of these components by means of a microelectrode technique gave the following results: (1) tetrodotoxin (TTX) abolishes the fast component of the upstroke; (2) the block of the fast component by TTX does not result in quiescence as the upstroke becomes entirely due to the slow component; (3) if excitation is elicited at higher take-off potentials, the fast component is enhanced in absence of TTX and does not reappear in the presence of TTX; (4) acetylcholine increases the take-off potential and therefore the fast component, while at the same time decreases the slow component; (5) in the presence of TTX, acetylcholine suppresses the slow response altogether; (6) increasing [Na]o in the presence of TTX enhances the magnitude of the slow response; (7) manganese decreases the slow component; (8) increasing [K]o leads to the complete substitution of the fast component by the slow component in the upstroke. It is concluded that excitation in subsidiary pacemakers of the sinus node is due to the activation of both a fast sodium channel (fast component) and a slow channel (slow component). Furthermore, one channel can entirely substitute for the other in that excitattion in the same cell is brought about through one channel when the other becomes inoperative." @default.
