- W2014332087 abstract "1. Rough-surfaced vesicles, smooth-surfaced vesicles and free ribosomes were isolated from rat liver at different times up to 30 min after injection of [14C]leucine. The rough-surfaced vesicles showed a higher incorporation of radioactivity at all time intervals after injection. 2. When the three microsome subfractions were separately incubated with [14C]leucine in the presence of cell sap pH 5 enzyme and an energy-generating system, the rough-surfaced vesicles had a higher capacity for amino acid incorporation per mg RNA in the preparation. 3. The free-ribosome fraction contained only 62-S and 83-S particles and showed amino acid incorporation in vitro that was independent of particle concentration. It is suggested that the free-ribosome fraction owes its activity to the presence of some active monosomes (single ribosomes coded with a strand of messenger RNA). 4. When rats were injected with [14C]adenine or 32P, the RNA of the free ribosomes showed a higher initial uptake of label than that of rough-surfaced vesicle ribosomes. This was shown to be due to a difference in labelling of both the 29-S and 18-S structural RNA of the free and attached ribosomes. It has been concluded that new ribosomes first enter the free-ribosome pool and are then continuously exchanged for ribosomes attached to polysomes in the membranes of the endoplasmic reticulum." @default.
