- W2014396460 abstract "In a recent communication (Ferguson, 1939) the results of experiments performed in an attempt to grow in sterile cultures the adult stage of a strigeid, Posthodiplostomum minimum (MacCallum, 1921), were briefly outlined. The present paper will show that, with this trematode of warm-blooded vertebrates, growth and tissue differentiation, resulting in the development of adult worms producing eggs and spermatozoa, have been obtained under controlled temperature conditions in artificial media free from micro6rganisms. No attempt is made here to review work of the several investigators who have tried to culture in vitro parasitic worms of vertebrates. The publications of McCoy (1935), Lapage (1937), Stunkard (1937), and Hoeppli, Feng, and Chu (1938) emphasized the desirability of growing these forms in artificial media, but none of the authors reported marked growth and tissue differentiation of parasitic worms in cultures. Glaser and Stoll (1938a, 1938b), however, made definite progress in the sterile cultivation of stages in the life cycle of the sheep nematode, Haemonchus contortus. They were able to carry their worms in cultures from the egg through several of the regular stages in development and to demonstrate growth and tissue differentiation. Using artificial media, Ackert, Todd, and Tanner (1938) obtained an increase in length in individuals of the fowl nematode, Ascaridia lineata, which had already begun their parasitic development in the intestine of chickens. Callot (1939) reported some development of the reproductive organs of the strigeid Posthodiplostomum cuticola when excysted metacercariae were incubated for some days in a physiological salt solution. Recently, Stunkard (1940) has referred to experiments in which metacercariae of Plagitura parva (adult stage in newts) were reared to sexual maturity in a bacteria-free artificial medium containing living yeast cells.
