- W2014513142 abstract "Abstract The dopamine (DA) hypothesis of schizophrenia, based on the prompt and striking remission of many schizophrenic symptoms to DA blocking drugs, suggests that DA or DA-like transmission in the brain is pathologically augmented in schizophrenia. Clinical evidence strongly favors a locus of that disturbance in one or more regions of the limbic striatum. There are a number of significant differences in the anatomy and pharmacology of the mesolimbic system compared with the nigro-striatal pathway; opioids, neuroendocrine agents and GABA exert a different influence on DA transmission in this pathway. However, high concentrations of GABA and GAD at several levels in the DA pathway permit opposing responses of GABA to occur, a fact which may explain the relative ineffectiveness of GABA agents in the treatment of schizophrenia. While GABA may inhibit DA neurons in substantia nigra-ventral tegmental area, GABA neurons may act synergistically with DA on striatum by depressing acetylcholine interneurons. Although there is conflicting evidence regarding abnormal GAD or GABA levels in the brains of schizophrenic patients and little evidence that treatment with putative GABAergic drugs exerts a favorable effect on these psychoses, ubiquity of the GABA network requires that GABA, GABA binding, or GAD in schizophrenic brains be measured in very specific regions of the mesolimbic DA pathway before the GABA hypothesis is rejected." @default.
