- W2014712312 abstract "We previously reported that (S,S)-formoterol enhances the production of IL-4, histamine, and PGD2 in mast cells (MCs) and promotes the inflammatory changes in the asthmatic airways. We thus hypothesize that MCs promote airway inflammation in response to (S,S)-formoterol. We determined the effect of formoterol isomers on MC-dependent airway inflammation using a murine model of asthma. MC-deficient WBB6F1-W/Wv (W/Wv) mice and wild-type (WT) mice received ovalbumin (OVA) with or without formoterol stereoisomers at a physiologically relevant concentration of 2.4 mg/kg by nebulizer for up to three weeks. Control mice received normal saline. Twenty-four hours after the last inhalation challenge with OVA, bronchoalveolar lavage fluid (BALF) and lung tissue were collected. To assess the inflammatory cellular infiltration to lung tissue, hematoxylin and eosin staining was performed. To determine the degree of airway inflammation in the airways, total cell numbers in BALF were counted. To determine the differential cell count, including eosinophils, MCs, and goblet cells, BALF cells were stained with Wright-Giemsa. We demonstrated that the degree of airway inflammation is significantly blunted in MC-deficient mice compared to WT mice, as indicated by the fewer inflammatory cells and MCs (P < 0.001) and that (S,S)-formoterol increases the number of eosinophils more in the lungs of WT than MC-deficient mice after OVA challenge (35% vs 23%; n = 5, P < 0.01). These findings suggest that MCs mediate the pro-inflammatory effect of (S,S)-formoterol in the asthmatic lungs." @default.
