- W2014758849 abstract "Studies have been carried out on the mechanism of inactivation of pyridine coenzymes in rat-liver mitochondria in the presence of inorganic phosphate and other agents. The inactivation has been found to occur through three stages: (a) the oxidation of the reduced coenzymes, primarily reduced TPN, (b) the leakage of the oxidized pyridine coenzymes from the mitochondria, and (c) the cleavage of the oxidized forms by an extra-mitochondrial DPNase. A correlation has been established between the oxidation of the reduced coenzymes and the swelling of the mitochondria. Substances which inhibit the swelling also suppress the oxidation. The destruction of the pyridine nucleotides is a secondary effect and is not related to the swelling process. Nicotinamide which inhibits the destruction of the coenzymes has no effect on the oxidation, release of the pyridine nucleotides or swelling. Succinate has been found to inhibit the release of the pyridine coenzymes as well as the swelling of the mitochondria. Malonate abolishes this effect of succinate. In addition to succinate, isocitrate and glutamate as well as cyanide and amytal are capable of suppressing the loss of the pyridine nucleotides from the mitochondria. Reduced coenzymes have not been observed to be lost directly from the mitochondria. Oxidation of the reduced forms appears to be essential before their release and destruction." @default.
