- W201498466 abstract "Students investigate the structure and function of the drug-metabolizing enzyme CYP2B4 or the signaling protein MAPK by site-directed mutagenesis (SDM). A cytochrome b5 expression system is being adapted for use, and has advantages of robust expression and easy methods of heme incorporation into apoprotein. Students visualize three-dimensional structure and then propose amino acid residues in the protein to alter by SDM. Instruction manuals and primary literature are used to perform SDM, and students confirm mutations by DNA sequencing. Results of student surveys in three laboratory courses in the Biology and Chemistry departments using inquiry-based projects reveal that 90% of the students gained a better appreciation for using online databases, for maintaining a lab notebook, for applying tools to address research questions, and for laboratory multi-tasking; over 80% received a better appreciation for understanding technical aspects and theory of methods, the benefits of biological model systems, and problem-solving; and over 70% had a better appreciation for writing scientific papers and of how research is conducted. Results support a positive effect of inquiry-based learning in the classroom. A website (www.emudnaprofiledatabase.org) was developed for one course for submission and analysis of Alu-based DNA profiles for multi-institutional participation.
