Under optimal conditions there is strict proportionality between amount of inoculum and resulting plaque count, the latter being affected by the volume and composition of the medium. The ratio of pock count on the chorioallantoic membrane to plaque count is usually less than 2:1. The amount of adsorption depends on concentration of virus, time, and temperature; adsorption occurs at 4°. After adsorption virus progressively becomes insusceptible to antiserum over a period of hours. The rate of this process is affected by temperature and does not occur at 4°. New virus from primarily infected cells spreads at first directly from cell to cell, but in cultures incubated for several hours it spreads through the medium as well." @default.
