Pronounced tissue changes, however, were observed during the first 3 weeks after the start of hormonal treatment. The total weight of the glands more than doubled during this period while the protein/DNA ratio decreased. Glucose- 6 -phosphate dehydrogenase (G- 6 -PDH) activity rose by 25% during the first 2 weeks of hormonal treatment calculated on a protein basis, but fell again to the initial value during the remainder of the induction period. Lactose synthetase (LS) A-protein activity and B-protein content rose to a steadily high level after 2 weeks of treatment. The non-blocked cytoplasmic estradiol binding capacity decreased after hormonal treatment of spayed virgins. In the induced tumors, the RNA/DNA ratio was unchanged and the protein/DNA ratio decreased by 25% compared to non-tumorous mammary tissue from the same mice. The activity of G- 6 -PDH increased by 50% calculated on a protein basis. The increased LS A-protein activity remained unchanged, while LS B-protein disappeared. There was 10 times as much non-blocked cytoplasmic estradiol receptor protein as in the non-tumorous tissue from the same animals.
