- W2016048092 abstract "SummaryThe anticholinergic effects of the tetracyclic antidepressant mianserin (10 mg), the tricyclic amitriptyline (25 mg) and a placebo were studied in a double-blind, randomized, group-comparative trial in 18 healthy young male volunteers. The daily dose of the test preparations was increased gradually from 1 tablet to the full level of the accepted antidepressant dose by Day 6 and maintained until Day 8. Measurements of various parameters indicative for anticholinergic effects were made before medication and on trial Days 2, 4, 6 and 8. Analysis of co-variance showed mianserin to be devoid of significant anticholinergic effects on any of the variables studied. It caused an un-explainable reduction in pupil diameter, tended to increase the distance of the near point of accommodation and increased salivary production as compared with placebo. Amitriptyline reduced saliva production significantly, tended to reduce pupil diameter and increased the distance of the near point. The subjective side-effects caused by actions on the autonomic nervous system were pronounced with respect to frequency and severity in the amitriptyline group and almost negligible in the mianserin-treated subjects. The trial demonstrates objectively that mianserin is practically free of atropine-like actions, whereas such effects are pronounced with a tricyclic antidepressant." @default.
