- W2016163661 abstract "버즈풋 트레포일(Lotus corniculatus L.) 및 이탈리안 라이그라스(Lolium multiflotrum L.) 의 치사온도를 결정하기 위하여, 각각 Au Dewey와 국내 육성품종인 화산 101호 품종을시험재료조 하여 종자를 Petri dish에서 발아시켜 작은 화분에 10 개체씩 이식, 생장실에서 4주간 재배하였다. 45, 50 및<TEX>$ 55^{circ}C$</TEX>에서 처리한 경우에는 60분간 처리했을 때에도 거의 식물체손상이 없었다. <TEX>$60^{circ}C/60$</TEX>분 처리에서는 잎이 약간 시든 듯한 현상이 있었으며, <TEX>$65^{circ}C/60$</TEX>분 처리에서도 식물체의 심한 손상은 나타나지 않았다. <TEX>$70^{circ}C/60$</TEX>분 처리에서 BFT의 일부에서 잎이 조금 마른 현상이 나타났고, IRG는 줄기 이외의 잎 부분이 거의 마른 상태가 되었지만 치사 온도에는 도달하지 않았다.<TEX>$ 80^{circ}C$</TEX>에서 처리한 BFT는 처리 후 1일 이내에 60분 처리에서 잎이 심하게 시든 현상이 나타났다. 처리 후 6일에는 60분 처리에서 모두 죽었으며, 55분 이하의 처리에서는 새로운 shoot가 재생됨을 확인하였다. <TEX>$80^{circ}C$</TEX>에서 처리한 IRC는 처리 후 1일 이내에 20분 이상 처리에서 거의 죽어가는 현상이 나타났다. 처리 후 7일에는 20분 이상의 처리에서 모두 죽었으며, 15분 이하의 처리에서는 심한 영향을 받지 않았으며 새로운 shoot가 재생됨을 확인하였다. 결론적으로 버즈풋 트레포일의 치사온도는 <TEX>$80^{circ}C/60$</TEX>분, 이탈리안 라이그라스의 치사온도는 <TEX>$80^{circ}C/20$</TEX>분으로 결정되었다. 다른 연구에서 형질전환 버즈풋 트레포일 및 이탈리안 라이그라스를 만들 경우, 형질전환체의 고온 내성을 간단하게 검정하는 방법으로 이상의 치사온도 결과를 이용할 수 있을 것으로 사료된다. To determine lethal temperature of birdsfoot trefoil(BFT) and italian ryegrass(IRG) at heat-stressed conditions, seedlings grown in a small pots fur 4 weeks were subjected to different temperature regimes of heat treatment. No apparent damage was observed BFT and IRG were treated at 45, 50 or <TEX>$60^{circ}C$</TEX> for 1 h. And also heat treatments at 60, 65 and <TEX>$70^{circ}C$</TEX> for 1 h, both of them were withered and showed damage symptom on their leaves but it was not lethal conditions for the whole plants. By contrast, most of plants were prominently withered within one day after heat treatment at <TEX>$80^{circ}C/60min$</TEX>. When BFT was exposed to <TEX>$80^{circ}C/60$</TEX> min, they were died within 6 days but there was found that new shoots were regenerated from the plants that had been treated at <TEX>$80^{circ}C$</TEX> within 55 min. IRC was also died within 2 days that exposed to <TEX>$80^{circ}C/20$</TEX> min but there was found that new shoots were regenerated from the plants that had been treated at <TEX>$80^{circ}C$</TEX> within 15 min. These results indicate that heat killing temperatures of BFT and IRG plants are <TEX>$80^{circ}C/60$</TEX> min an <TEX>$80^{circ}C/20$</TEX> min respectively. Simple viability assay system established in this study will be useful for selection and characterization of heat-tolerant transgenic BFT and IRG plants." @default.
