- W2016273672 abstract "Abstract Wheat striate mosaic virus (WSMV) was extracted from experimentally infected Ramsey wheat (Triticum durum Desf.) using a Tris-HCl buffer containing 0.15 M d -mannitol and 0.001 M ethylenediaminetetraacetic acid (EDTA), pH 8.0. After clarification of the extract by low-speed centrifugation, virus was sedimented at 26,000 g for 20 minutes and examined in the electron microscope by the negative staining technique. The virus is bacilliform (176 × 78 mμ) appears flat at one end and consists of an outer membrane 5 mμ in width enclosing a dense core with a central hollow cylinder 23 mμ wide. Infectivity of the virus was determined by injecting it into virus-free leafhoppers (Endria inimica Say.), then testing the injected insects on healthy wheat seedlings. In two assays, the percentage transmission obtained using extracts containing bacilliform particles was 43% and 41%, respectively. Localization of WSMV was determined by studying thin sections taken from plants at various intervals after infection. Virus was found in both parenchyma and phloem companion cells, and particles were localized in the cytoplasm, between nuclear membranes, and in nuclei of infected cells." @default.
