- W2016397721 abstract "Abstract A system of yeast ilv1 mutants, well-characterized by intragenic complementation and allelic mapping, was used for suppressor studies. The ilv1 locus of Saccharomyces cerevisiae is the structural gene for threonine dehydratase (EC 4.2.1.16). The enzymatic activity is sensitive to feedback inhibition by L -isoleucine. Conditional and complementing ilv1 mutants and the effect of their suppressors were identified by genetic and biochemical procedures. A nonsense (UAA) suppressor was determined for the temperature-sensitive mutant ilv1-113. Two missense suppressors were analysed from osmotic-remedial and intragenic-complementing ilv1 alleles. None of the isolated, spontaneous revertants was a true reversion to the wild type and only one intragenic suppressor was found. It is suggested that the allele-non-specific but locus-specific suppressors of osmotic-remedial mutants act by opening an alternative pathway for the synthesis of isoleucine. There was no pleiotropic effect of the missense suppressors on the allosteric regulation of threonine dehydratase and isocitratelyase (EC 4.1.3.1) or growth, which could be the result of suppressor mutants coding for a highly redundant tRNA species." @default.
