- W2016522306 abstract "We report on the engineering of hardware systems and molecular tools to allow the rapid detection and discrimination of sub-populations of Ralstonia solanacearum, a bacterial wilt pathogen affecting a wide host-range of agronomically important plants, which also exhibits a large degree of virulence and environmental fortitude within the species. The hardware is based on a compact disc platform, wherein DNA delivered to a reaction well is replicated at a constant controlled temperature, and detected in real-time through increased fluorescence caused by the displacement of a quencher strand from a fluorescent “molecular zipper” during the DNA polymerization. The primers designed for the reaction were shown to selectively replicate DNA from race 3 biovar 2 populations of the pathogen, which is considered a major threat to potato growers in North America if it were ever introduced, but not DNA from other strains of Ralstonia solanacearum. Selective replication was observed in real time using the molecular zippers. Work is ongoing to demonstrate the molecular replication and detection on the CD media, though results have demonstrated that the temperature control on the CD is effective using non-contact measurements corrected for transmission losses in the reaction well and reflection of ambient radiation. Ultimately, we intend to integrate all of these systems onto a CD where centrifugal control can be used to sequence sample capture, wash, lysis, and DNA extraction followed by the replication and detection on an inexpensive disposable media." @default.
