- W2016958616 abstract "Cumulus expansion occurs as a result of the preovulatory gonadotropin surge. Cumulus-oocyte complexes (COC), isolated from mice, expand in vitro in the presence of FSH or PGE2. Endogenous PGE2 supplied by ovarian COX-2 mRNA, has been shown to be expressed 4 h after hCG stimulation in rats. The aim of the present study was to evaluate the effect of PGE2 and a receptor selective PGE2 analog (AS02) added to IVM 5 h after the beginning of the IVM on subsequent embryo development. Oocyte quality after IVM was evaluated by fertilization and blastocysts rate. AS02 was developed at SRBI by rationale drug design. Adult B6CBAF1/J females were primed with 5 IU of PMSG. Oocytes were collected 48 hs later. The oocytes were matured, fertilized and cultured in vitro during 5 days. The rate of embryo development up to blastocyst was recorded in 6 groups: 1) Oocyte matured with αMEM plus Fetal Calf Serum (IVMm)[Negative Control]; 2) IVMm plus 0.2 IU rFSH (IVMmfsh)[Positive Control]; 3) IVMm plus 1 μM PGE2; 4) IVMm plus 1 μM AS02; 5) IVMmfsh plus PGE2 or 6) IVMmfsh plus AS02. Logarithmic transformed data were analysed by ANOVA and Tukey’s test. Results are expressed as Mean ± Sdv. Cleavage rate and blastocyst rate were significantly increased when PGE2 was added to IVMmfsh when compared with IVMmfsh alone (cleavage: 90 ± 7 % vs. 61 ± 4 %; Blastocysts: 79 ± 2 % vs. 61 ± 4 %, respectively P<0.05). When AS02 was added to the IVMmfsh, cleavage rate, but not blastocyst rate, was significantly increased when compared with IVMmfsh alone (Cleavage: 81 ± 7 % vs. 61 ± 4 %; P<0.05, Blastocysts: 69 ± 3 % vs. 61 ± 4 %, respectively, P>0.05). Moreover, PGE2 when added alone to IVMm had similar cleavage and blastocyst rate as IVMm plus FSH (IVMmfsh) (Cleavage: 60 ± 10 % vs. 61 ± 4 %; Blastocysts: 57 ± 14 % vs. 61 ± 4 %, respectively). In comparison to IVMm containing only PGE2 or FSH, addition of AS02 to IVMm did not increase cleavage rate or blastocyst rate (Cleavage: 19 ± 11 %, Blastocysts: 11 ± 7 %). In summary, in the presence of FSH, PGE2 and AS02 significantly improved oocyte quality as measured by cleavage rate but only PGE2 induced an increase on blastocyst development. These data demonstrate that the effects of a receptor selective analog of PGE2 mimics some but not all of the effects of natural PGE2. Furthermore, the combined presence of FSH and PGE2 during IVM significantly increased developmental rates." @default.
