- W2017007250 abstract "Crude lipase was extracted from defatted ground French bear seeds using 1 × 10 −3 M CaCl2 in combination with ultrasound disruption and further purified with ammonium sulfate at 60% of saturation. The additional use of ammonium sulfate doubled the crude lipase activity, while the combination of ultrasound and ammonium sulphate increased the activity by a factor of six. The effects of pH, calcium ion concentration and the addition of an emulsifying agent were also investigated. The optimum pH was determined to be near 7, and the addition of calcium ion beyond the concentration present in the extract had an inhibitory effect on the activity, whereas the addition of Tween-20 increased the activity of the enzyme by a factor of four. The specificity of French bean lipase, relative to pancreatic lipase, was studied using triacetin, olive oil and French bean lipid as substrates. The crude lipase fraction obtained from the French bean seed had a higher activity than pancreatic lipase and it also exhibited notable carboxulesterase activity. La lipase fut ensuite purifiée au sulfate d'ammonium saturé à 60%. L'addition d'ion calcium au-delà de la concentration présente dans l'extrait eut un effet inhibitoire sur l'activité, tandis que l'addition de Tween-20 augmenta l'activité de l'enzyme par un facteur de quatre. La spécificité de la lipase d'haricots verts fut étudiée en rapport avec la lipase pancréatique à l'aide de triacétine, d'huile d'olive et de lipides d'haricots verts comme substrats. La lipase d'haricots verts fut plus active que la lipase pancréatique et, de plus, elle montra une activité carboxylestérasique importante." @default.
