- W2017010723 abstract "Abstract Turkish tobacco cells infected with two defective mutants, PM1 and PM2, of tobacco mosaic virus (TMV) were examined cytologically. Parallel observations were made on symptoms, variability, and infectivity of invaded tissues. Symptoms observed were much like those originally described, but symptom variants were readily obtained by transfers from single lesions. There was occasional evidence of erratic systemic translocation of virus. Concentration of virus was high in infected tissues before symptoms appeared, low in the senescent chlorotic tissues of old lesions, but higher at their edges. No infectivity was found in necrotic tissues. Cells long infected with PM1 contained a network of gelled (virus) material consisting mainly of a component staining like virus protein. Dyes that gave selective staining of RNA showed that RNA was also present in the gel. It was removable by ribonuclease without apparent diminution of the background material. Protein material of PM2 was aggregated in longitudinally striate needles and loops. The RNA was largely excluded from these aggregates, and appeared in the cytoplasm as “X-body material” like that in cells infected with the parent strain, TMV-U1. In the early stages of infection RNA appeared to originate in the nucleolus and pass through the nucleus to the cytoplasm, as in the parent strain, TMV-U1. In living cells under phase optics this process was associated with emission of optically clear material from the nucleolus. Evidence was seen of another form of emission from the nucleolus. Material optically more dense than the nucleoplasm seemed to be given off from a nucleolar projection in repeating cycles of less than 5 minutes. In stained preparations of what may have been the same sequence, no RNA staining of the material given off was obtained.
