- W2017923995 abstract "It is known that in cultured cells treated with low doses of toyocamycin (1 μg/ ml) and less, there is a selective inhibition of the cleavage of the 45 S nucleolar RNA synthesized in the presence of the drug. This RNA species accumulates in the nucleoli, whereas the 45 S RNA synthesized prior to the administration of the drug is normally processed. This study demonstrates that in the nucleoli of treated cultured kidney cells, there is no obvious accumulation of RNP fibrils nor loss of granules, a fact which correlates with the finding that at least some of the granules are associated with RNAs heavier than 28 S or 18 S. Higher doses of toyocamycin (5 μg/ml) largely inhibit overall RNA synthesis, and induce a spotted appearance of the nucleoli, which become uncoiled and fragmented within 30 minutes. High-resolution autoradiography reveals an important decrease in RNA synthesis in the nucleoli, the only labeled parts of which are the fibrillar rods and the spots. Although cultured cells treated with millimolar amounts of adenosine do not stop RNA synthesis, they do not incorporate the added tritiated uridine and they display nucleolar alterations very similar to those given by lower doses of toyocamycin. The resemblance of these lesions to those induced by ethionine in liver cells suggest that disturbances in the nucleotide pool may be a possible common basis for the phenomenon of nucleolar fragmentation." @default.
