- W2018306730 abstract "Abstract Fluorescence properties of the red phytochrome form (Pr) in etiolated oat seedlings and 124-kDa oat phytochrome in vitro (A666/A280=0.93) (Pr′) were directly compared for the first time. Using a number of parameters, Pr′ was found to be quite close to Pr. The maxima were located in the low-temperature (85 K) emission spectrum at 685 nm and 684 nm, and in the excitation spectrum at 673 and 672 nm for Pr and Pr′, respectively. With the rise in temperature from 85 K to 293 K the fluorescence yield (ϕF) decreased 30 – 40 times in both cases. The decreases in fluorescence connected with the Pr⇄Pfr phototransformation are also practically identical, 0.80 – 0.82. However, other pigment characteristics undergo changes after its isolation. There are differences in the ϕF temperature dependence and, correspondingly, in the activation energy (Ea) of the fluorescence decay; in Pr Ea = 3.9 – 4.1 kJ mol−1 at temperatures of 218 K and below and Ea = 23.9 kJ mol−1 at 218 K and above. Within the mentioned temperature intervals Ea is 2.7 – 3.0 kJ mol−1 and 17 – 20 kJ mol−1 for Pr′. The variable fluorescence connected with irreversible and reversible phototransformations of the pigment into the photoproduct (lumi-R) at low temperatures (below or equal to 200 K) was 2 – 3 times lower in Pr′ than in Pr. The data are interpreted in terms of several native species of phytochrome whose relative concentrations and, probably, properties undergo some changes during isolation of the pigment." @default.
