- W2018464320 abstract "Abstract A method is described for determining the affinity constants and antibody concentrations that give the best fit to a hapten-antibody binding curve obtained for an equilibrium involving one hapten and several antibodies. The best estimates of the above parameters are obtained by means of a non-linear least-squares minimisation procedure which at each step calculates the hapten-antibody complex concentrations using a Newton-Raphson iteration. A method of estimating the individual quenching constants, if the binding curve is derived from fluorescence quenching data, is also described. As the binding equations are non-linear with respect to the unknown parameters, there is no rigorous method of determining which model, i.e. how many antibodies give the best fit or for calculating the standard deviations of the estimated parameters. However, approximate methods for deciding on the former and for setting a lower bound, the Crame´r-Rao inequality, on the latter are described. An extensive set of data describing the binding of DNP-aminohexanoate to mouse anti-DN P antibody is analysed in detail in these terms." @default.
