- W2019437866 abstract "Callus cultures originating from orange and lemon flavedo were established and their successful growth was maintained through several subcultures. Best growth was obtained with Murashige and Skoog's medium containing high levels of sucrose (5 %), B-vitamins (10 times original concentration), IAA (10 ppm) and kinetin (1 ppm). Conconut water was required for the initiation of callus tissue and its continued growth. Orange callus tissue grown in the dark was characterized by profuse growth and the absence of cell differentiation and lignification. Its phenylalanine ammonia-lyase (PAL) activity increased slightly after 48 hours from subculture, and remained almost constant at a low level for the rest of the growth period. Trace amounts of flavonoids were formed in dark-grown tissue. Exposure of the subcultured callus to light (ca. 3000 lux) resulted in active cell division, tissue differentiation and extensive lignification. There was an 8-fold increase in PAL activity after 24 hours of illumination and a distinct maximum was reached after 48 hours. This was followed by a rapid decrease in enzyme activity. Flavonoid production was evident in the light-grown tissue, with sinensetin and nobiletin being the major components. However, the amounts of flavonoids formed in the callus were relatively low as compared to those found in the peel expiants." @default.
