- W2019504384 abstract "The changes in morphology of Saccharomyces cerevisiae during freezing and thawing were observed cryomicroscopically. With cells from a late exponential phase culture at 24 °C there was a correlation between the morphology during freezing and viability on thawing. At the optimum rates of cooling for viability (3 to 10 °C−1), cells shrink extensively during freezing. At greater rates of cooling the reduction in viability was associated with the formation of intracellular ice. At suboptimal rates of cooling, cells were osmotically unresponsive, indicating damage to the cell envelope. Cells obtained from the late exponential phase of culture at both higher and lower temperatures were more resistant to freezing injury. At any temperature of culture, stationary phase cells were more resistant to freezing than those from the late exponential phase. After these alterations in growth conditions, cells were extremely resistant to hypertonic stress and retained viability when intracellular ice nucleated." @default.
