- W2019697958 abstract "Thyroid iodoprotein frozen in a solution of methimazole (MMI) was more readily hydrolyzed by a dilute solution of pancreatin after subsequent thawing than when frozen in the absence of MMI. This phenomenon was observed in labeled iodoprotein from intact iodine-replete rats and intact and hypophysectomized iodine-deficient rats. Addition of MMI without freezing did not alter the rate of proteolysis. Concentrations of MMI of 0.005M or less were ineffective. An increased rate of proteolysis in the MMI-prefrozen iodoprotein was not due to release of cofactors or proteolytic enzymes from the frozen tissue. Mercaptoethanol, an agent known to disrupt disulfide bonds, increased the rate of proteolysis of iodoprotein when added to the solution at room temperature. There was no potentiation of the mercaptoethanol effect by freezing, nor was there an additive effect on the rate of proteolysis by combining mercaptoethanol pretreatment and freezing in MMI There was a decrease of approximately 25% in the yield of label..." @default.
