- W2019927351 abstract "Relatively mature red cell precursors (CFU-E) from freshly explanted mouse bone marrow proliferate and differentiate to form colonies of erythroblasts in culture under the influence of erythropoietin. This process normally depends on the presence of serum in the medium. Serum can be replaced entirely by the combination of delipidated albumin, transferrin, iron, and sonicated mixtures of unsaturated fatty acid, phosphatidyl choline (lecithin) and cholesterol. Plating efficiency of CFU-E in the serum-free system consistently equals that obtained with high serum concentrations. The erythropoietin dose-response curve in the new conditions is similar to that with serum except that background colony formation is more reliable at or near zero. Colony formation remains linearly related to seeding density down to limiting dilutions of CFU-E. Highly purified erythropoietin (70 000 U/mg) is fully active without serum. The results emphasize the stringency of the erythropoietin requirement for adult CFU-E, suggest that other serum hormones are not required for the terminal differentiation events, and provide a system in which the effects of accessory factors can be studied free of the confusion contributed by serum." @default.
