- W2019977553 abstract "The mode of formation of ketone bodies from butyrate by tissue from the rumen and omasum of the sheep has been investigated by the use of four types of butyrate labelled separately in each of its carbon atoms. When [1-14C]butyrate or [3-14C]butyrate is metabolized, there is an equal isotope distribution between carbon atoms one and three of the ketone bodies. A similar distribution to that produced by [1-14C]butyrate is found when small amounts of labelled compounds which are metabolized via [1-14C]acetyl CoA are incorporated into ketone bodies in the presence of unlabelled butyrate. From the results on this isotopic distribution it has been calculated that about three-quarters of the butyrate carbon chains are converted to ketone bodies without a breakdown and recombination by the thiolase reaction. Of the ketone bodies formed in the presence of butyrate, about three-quarters have their origin in the carbon atoms of butyrate which have therefore undergone appreciable dilution by carbon atoms of endogenous origin. Two to four times as many carbon atoms one and two of butyrate are oxidized to carbon dioxide via the tricarboxylic acid cycle as carbon atoms three and four. The results are interpreted on the basis of a scheme for the conversion of acetoacetyl CoA to acetoacetate which is considered to operate in liver, omasum and rumen tissue and which very strongly suggests the operation of the β-hydroxy-β-methylglutaryl pathway in the synthesis of acetoacetate." @default.
