En effet, toute variation de leur concentration plasmatique sera interprétée par l’organisme comme un manque ou un excès d’énergie. En ce qui concerne les acides gras libres (AGL), il est à présent reconnu que les variations périphériques de concentration des AGL peuvent être détectées par des neurones spécialisés, notamment dans l’hypothalamus et le tronc cérébral. Ainsi, les AGL régulent finement, via leur effet central, le comportement alimentaire, la production hépatique de glucose mais aussi la sécrétion d’insuline. Des sous-populations de neurones sensibles (inhibés ou excités) aux AGL sont identifiées dans l’hypothalamus. Leur caractérisation moléculaire reste encore à établir. Plutôt que les AGL directement, il semble que ce soit leurs métabolites (acyl-CoA et/ou le malonyl-CoA par exemple) qui sont les principales molécules actives. Par exemple, une élévation de la concentration intracellulaire de malonyl-CoA est un puissant signal satiétogène. Physiopathologiquement, un apport excessif chronique d’AGL pourra entraîner un dysfonctionnement du senseur central des AGL et contribuer, chez les sujets prédisposés, à l’apparition de maladies métaboliques, telles que l’obésité ou le diabète de type 2. Free fatty acids (FFA) are not fuel for neurons and do not cross “easily” blood brain barrier. However, it is know well established that FFA could act as informative molecules and any change in plasma FFA concentration are detected at the level of central nervous system (CNS) structure such as hypothalamus and brainstem, where FFA sensitive neurons are located. These neurons control in turn energy homeostasis including food behaviour, hepatic glucose production or insulin secretion. Sub-populations of FFA sensitive neurons (either excited or inhibited) are now described in hypothalamus. Regarding, cell signaling, both acyl-CoA and malonyl-CoA could be major factor to mediate FFA effect. Indeed, increased intracellular concentration of malonyl-CoA in hypothalamus induce food intake inhibition. Such effects are partly related to decrease in NPY gene expression, as well as increase in alpha-MSH expression. Finally, dysregulation of FFA sensing in CNS could lead to development of metabolic diseases such as obesity or type 2 diabetes, in predisposed subject." @default.
