The pH was found to be a suitable indicator for the optimum harvest time. Cell wall permeabilization by Triton yielded β-galactosidase activity of up to 966 LAU (g cell) -1 . The effect of Triton treatment was documented by SEM pictures of the cells. The cells were immobilized in polyurethane foam and used in a continuous plug-flow reactor with a half-life time of approximately 3 days. It was shown that both the free as well as the immobilized cells utilize glucose in a follow-up reaction and, therefore, kinetic measurements based on glucose concentrations alone are not sufficient to evaluate true kinetic parameters." @default.
