- W2020174164 abstract "A simple and rapid radiochemical method is described for the assay of adenine and hypoxanthine/guanine phosphoribosyltransferases activities. After the incubation of enzyme with (8-14C) adenine or hypoxanthine, Mg++ and 5′-phosphoribosylpyrophosphate, the labelled nucleotide products are isolated by selective elution from small alumina column. Adenine, hypoxanthine, adenosine, xanthine and uric acid are eluted from column by 0.1 M acetate buffer (pH 4.0) or 0.1 M acetic acid. The nucleotide products, adenosine monophosphate and inosine monophosphate are subsequently eluted with 0.25M H3PO4. The method was utilized in the determination of the regional distribution of both adenine and hypoxanthine/guanine phosphoribosyltransferases activities in the rat brain. The highest activities of both enzymes were found in the hypothalamus and the lowest in the medulla plus pons. The major advantages of this method are a lack of interference by the products of xanthine oxidase action, an ease of handling, speed and reproducibility. Fifty to 100 assays are easily performed in a day." @default.
