- W2020289966 abstract "We previously identified by Southern blot analysis an mRNA that encodes a 26-kDa protein, DAM1 (DNA amplified in mammary carcinoma 1), which was amplified in the MCF-7 and BT-20 breast cancer cell lines. The amplificationlevels of the DAM1 gene were estimated to be 10- and 5-fold in MCF-7 cells and BT-20 cells, respectively, compared with that in the other examined breast cancer cell lines. Recently, the DAM1 protein was identified to have 100% homology to the sequence of a spliceosome associated protein (GenBank Accession No. AF0817889) by BLAST database search. In this study, we used the enhanced green fluorescent protein (EGFP) technique to determine the subcellular localization of the fusion protein, EGFP-DAM1. We generated a chimeric gene encoding the full-length DAM1 protein with the coding sequence of GFP fused to its C terminus. This construct was stably transfected into MCF-7 cells and the intracellular localization of the recombinant protein was studied by fluorescence microscopy. Whereas wild-type GFP was distributed throughout the cell, EGFP-DAM1 was localized predominantly in the nucleus. Although the function of DAM1 remains unknown, our present results suggest that the DAM1 protein has a specific nuclear role in some breast cancer cell lines." @default.
