- W2020768175 abstract "Background: The increases in influenza-related hospitalizations and mortality despite the availability of the influenza vaccine and antiviral drugs such as amantadine and rimantadine underscore the need for better treatments for influenza. Objective: This review describes the epidemiology, pathogenesis, diagnosis, and management of influenza with special focus on the neuraminidase inhibitor zanamivir. Results: Introduced in 1999 as the first neuraminidase inhibitor for the treatment of influenza A and B, zanamivir fills many of the therapeutic needs unmet by the older medicines. With a unique mechanism of action and a topical mode of delivery (inhalation), zanamivir is the only treatment available that offers broad-spectrum efficacy against both influenza A and influenza B combined with a favorable tolerability profile. The risk of developing clinically relevant resistance to zanamivir during normal clinical use appears, on the basis of data collected to date, to be minimal. No zanamivir-resistant strains were isolated in phase II or III clinical trials involving more than 4000 patients. Zanamivir is effective in the prevention of influenza as well as its treatment. The majority (71%) of 1408 patients returning surveys about their experience with zanamivir during the 1999 influenza season indicated that they were satisfied with their treatment. Conclusions: Zanamivir, an advanced over previous antiviral therapies for influenza, can aid physicians in efforts to reduce the impact of this debilitating disease." @default.
