- W2020792110 abstract "To study the potential effect of magnesium sulfate (MgSO4) and angiotensin II (AngII) on the levels of prostaglandin E2 (PGE2) in preeclamptic placentas. Cotyledons from term preeclamptic placentas were dually perfused. Rising doses of AngII were injected into the maternal reservoir of placentas perfused with MgSO4 (6-7 mg %) in the maternal reservoir [normotensive (n=3); preeclamptic (n=5)] or with control medium (without MgSO4) [normotensive (n=3); preeclamptic (n=5)]. In addition, preeclamptic placentas perfused with MgSO4 and injected with AngII were compared with placentas perfused with MgSO4 and not injected with AngII. Perfusate samples from maternal and fetal circulations were repeatedly collected during 3h of perfusion, and examined for PGE2 levels by radio-immuno assay (RIA). RIA results were normalized for gram of perfused cotyledon. Statistical significance was determined using 2-way analysis of variance (ANOVA). Significantly higher levels of PGE2 were observed in the fetal circulation of preeclamptic placentas after perfusion with MgSo4 and injection of AngII as compared to placentas following injection of AngII without perfusion with MgSO4 (p<0.005). In contrast, significantly lower levels of PGE2 were observed in the maternal circulation of preeclamptic placentas after perfusion with MgSo4 and injection of AngII as compared to placentas not perfused with MgSo4 (p<0.05) (figure). In both, fetal and maternal circulations of preeclamptic placentas that were perfused with MgsO4, higher levels of PGE2 were noted after injection of AngII, as compared to those not injected with AngII. MgSO4 increases the secretion of PGE2 to fetal circulation and decreases the secretion of PGE2 to maternal circulation in preeclamptic placentas after injection of AngII.
