- W2021009768 abstract "Chlorphencsin carbamate（CPC）を1～4週間連続投与し，その耐薬性の発現について薬効と血清内の遊離CPC濃度，肝のUDP-glucronyltransferase活性，cytochrome P-450含量の変化から検討し，つぎの結果を得た．マウスではhexobarbital睡眠時間および筋弛緩効果ともにCPCの7日間連続投与で減少し，14日目には回復する傾向を示したが，血清内の遊離CPC濃度の経日変化も薬効と同様のパターンで推移し，またこの期間薬効と血清内の遊離CPC濃度との間に有意の相関が得られた．ラットでは，薬効はマウスとほぼ同様の経日変化を示したが，肝の薬物代謝酵素活性は14日間の連続投与期間中いずれも対照値より高値を示し，それは血清内の遊離CPC濃度とよく一致した経日変化を示した．すなわちマウスにおける14日目の薬効の回復傾向は肝の薬物代謝酵素活性が回復過程にあたるためと考えられ，ラットにおいては回転棒へのhabituationによると考えられる．以上のようにCPCの薬効発現は血清内の遊離CPC濃度，薬物代謝酵素の活性に強く依存しており，CPCの連続投与では肝薬物代謝酵素の誘導により耐薬性が生じたと考えられる．" @default.
