- W2021027467 abstract "A new method of induction of tolerance to allogeneic and to xenogeneic cells is presented. It includes thymectomy of adult mice followed 1 month later by the injection of 1 × 108 spleen cells i.v. and i.p. administration of 200 mg of cyclophosphamide per kg 1 day after cells. This method induced prolonged survival of heterotopically transplanted neonatal C57BL/6 marine heart grafts (more than 8 months) and of August rat heart grafts (more than 2 months) in CBA mice. Tolerance to allo- or xenoantigens was formed at the cell level. Experimental animals did not produce allo- or xenohemagglutinins after graft implantation. Spleen cells of mice with surviving C57BL/6 heart grafts did not respond to C57BL/6 cells in mixed lymphocyte culture (MLC) reaction. Lymphoid cell chimerism was not observed in animals tolerant to alloantigens. Long-lasting chimerism of lymphoid or hemopoietic cells is known to prolong survival of allografts (1). Chimerism can be achieved by injection of a large dose of donor cells into animals treated with immunosuppressive agents such as X-rays, γ-irradiation, or certain drugs. Thus, injection of mice with semial-logeneic spleen cells several hours after cyclophosphamide (CY) administration provides long-lasting chimerism of lymphoid tissue and makes the animals tolerant to the donor skin or heart grafts (2–5). However, it is difficult to obtain lymphoid cell chimerism in allogeneic or xenogeneic combinations since donor cells generate graft-versus-host reactions in the immunosuppressed recipients. In this study, we attempted to establish long-lasting tolerance to allo- and xenogeneic cells in nonchimeric mice. Thymectomy of adult mice and subsequent injections of donor spleen cells and CY was used for this purpose. Pretreatment of mice with donor spleen cells and CY before allogeneic lymphocyte administration in a previous study (6) caused some prolongation of the survival of the transplanted cells. Thymectomy has been used by other workers with anti-lymphocyte serum and donor cell administration for prolongation of skin graft survival (1, 7–9). Combination of thymectomy, donor cell, and CY administration proved to be efficient in our studies in inducing prolonged survival of allogeneic or xenogeneic heart grafts." @default.
