- W2021332746 abstract "Tumor necrosis factor-alpha (TNF-alpha) is a pro-inflammatory cytokine involved in the pathogenesis of Alzheimer's disease (AD). Recent experimental studies suggest that TNF-alpha is a gliotransmitter acting as a regulator of synaptic functions and as a mediator in the disruption of memory mechanisms induced by beta-amyloid. There is also evidence that the signalling pathways of TNF-alpha and insulin-like growth factor I (IGF-I), a pleiotropic hormone, are functionally interrelated. It has been demonstrated that TNF-alpha counteracts the capacity of IGF-I to promote neuronal survival and antagonizes brain amyloid-beta clearance induced by serum IGF-I, whereas IGF-I reverses neuronal apoptosis mediated by TNF-alpha. According to the available scientific data we postulated that a hyperactivity of the peripheral TNF-alpha system as well as a negative interaction with serum IGF-I might be present in AD. Serum levels of TNF-alpha, soluble TNF receptors (sTNF-R I and II), total IGF-I, dissociable IGF-I, free IGF-I and IGF binding proteins (IGFBP-1, IGFBP-3) were evaluated in AD, mild cognitive impairment (MCI) and elderly control subjects (EC). Circulating TNF-alpha levels were found to be significantly (p 0.01) increased and to correlate negatively with dissociable-free IGF-I in AD patients (Álvarez et al, Neurobiol Aging, 2007). This correlation was not evidenced in control subjects or in MCI cases. A significant reduction (p 0.05) of age-adjusted total IGF-I, but not dissociable-free IGF-I levels was observed in AD as compared with controls. Serum levels of soluble TNF Receptor I and IGF-I binding protein 3, both having pro-apoptotic activity, were also found to be higher in AD patients than in controls (p 0.01 & p 0.05, respectively). However, sTNF- RII and IGFBP-1 showed no changes in AD sera. According to our results, it seems that a hyperactivity of the peripheral TNF-alpha system exists in AD patients and that the opposite influence of inflammatory (TNF-alpha) and growth (IGF-I) factors on AD pathology is having a peripheral reflect." @default.
