- W2021463352 abstract "Hydrochlorothiazide and amiloride have been used in combination to reverse the hypercalciuria induced by calcitriol (1,25-dihydroxyvitamin D3) administration. In order to evaluate the fate of the retained calcium under conditions of chronic diuretic use, forty-two male, weanling Sprague Dawley rats were randomly assigned to control and treatment groups for four weeks. The animals were fed laboratory rat chow. The hydrochlorothiazide at 3 mg/kg/day, amiloride at 0.6 mg/kg/day and calcitriol at 20 or 50 ng/kg/day were given by stomach tube. Our results demonstrated that with the concurrent administration of amiloride and calcitriol at conventional therapeutic dosages, the total calcium content of the whole kidney was decreased without affecting the femur calcium concentration. The hydrochlorothiazide-treated rats, compared with the amiloride-treated rats, had a higher concentration of total kidney calcium (206–213 versus 183–188 μg/g dry weight; p<0.05), with no difference in the femur calcium (171–182 versus 178–181 mg/g dry weight). The hydrochlorothiazide tended to decrease urine calcium in comparison with the amiloride-treated animals (2.14–2.82 versus 3.39–3.83 mg/24 hours). These results suggest that a possible relationship exists between kidney calcium content and tubular calcium transport with chronic use of amiloride with calcitriol." @default.
